Title: Getting the Right hPSC-derived Cardiomyocytes for the Job

Abstract: Human pluripotent stem cell-derived cardiomyocytes (hPSC-CMs) are a promising cell source for cardiac therapies and generating models of heart disease. However, robust methods to generate hPSC-CMs representing right ventricle (RV) are not available. Using signaling to mimic the development of the second heart field, we generate hPSC-rvCMs and compare them with other chamber-specific CMs using scRNA-seq, electrophysiology and Ca2+ cycling. The hPSC-rvCMs are used to model inherited heart disease which specifically can impact the right ventricle including Brugada Syndrome and Arrhythmogenic Cardiomyopathy. The studies further refine the ability of hPSC-CMs to investigate and develop therapies for the right-sided heart disease.
